ABSTRACT

This paper aimed to investigate the role and potential mechanism of p53 on primordial follicle activation. Firstly, the p53 mRNA expression in the ovary of neonatal mice at 3, 5, 7 and 9 days post-partum (dpp) and the subcellular localization of p53 were detected to confirm the expression pattern of p53. Secondly, 2 dpp and 3 dpp ovaries were cultured with p53 inhibitor Pifithrin-μ (PFT-μ, 5 μmol/L) or equal volume of dimethyl sulfoxide for 3 days. The function of p53 in primordial follicle activation was determined by hematoxylin staining and whole ovary follicle counting. The proliferation of cell was detected by immunohistochemistry. The relative mRNA levels and protein levels of the key molecules involved in the classical pathways associated with the growing follicles were examined by immunofluorescence staining, Western blot and real-time PCR, respectively. Finally, rapamycin (RAP) was used to intervene the mTOR signaling pathway, and ovaries were divided into four groups: Control, RAP (1 μmol/L), PFT-μ (5 μmol/L), PFT-μ (5 μmol/L) + RAP (1 μmol/L) groups. The number of follicles in each group was determined by hematoxylin staining and whole ovary follicle counting. The results showed that the expression of p53 mRNA was decreased with the activation of primordial follicles in physiological condition. p53 was expressed in granulosa cells and oocyte cytoplasm of the primordial follicles and growing follicles, and the expression of p53 in the primordial follicles was higher than that in the growing follicles. Inhibition of p53 promoted follicle activation and reduced the primordial follicle reserve. Inhibition of p53 promoted the proliferation of the granulosa cells and oocytes. The mRNA and protein expression levels of key molecules in the PI3K/AKT signaling pathway including AKT, PTEN, and FOXO3a were not significantly changed after PFT-μ treatment, while the expression of RPS6/p-RPS6, the downstream effectors of the mTOR signaling pathway, was upregulated. Inhibition of both p53 and mTOR blocked p53 inhibition-induced primordial follicle activation. Collectively, these findings suggest that p53 may inhibit primordial follicle activation through the mTOR signaling pathway to maintain the primordial follicle reserve.

ABSTRACT

AIM: To investigate the correlation between axial length and diabetic retinopathy(DR)in patients with type 2 diabetes mellitus.METHODS:This study is a cross-sectional study. A total of 53 cases(104 eyes)of type 2 diabetes patients who admitted to the ophthalmology department of the Jining No.1 People's Hospital between January and May 2023 were included. Among these cases, 51 patients had both eyes included, while 2 patients had only one eye included. The patients were divided into two groups based on the presence or absence of fundus lesions. The non-diabetic retinopathy(NDR)group consisted of 32 eyes, and the DR group consisted of 72 eyes. Within the DR group, the patients were further categorized based on the severity of fundus lesions. The non-proliferative diabetic retinopathy(NPDR)group comprised of 27 eyes, and the proliferative diabetic retinopathy(PDR)group consisted of 45 eyes. The axial lengths of eyes in the DR group were divided into four groups using quartiles: 19 eyes in the 20.00~22.09 mm group, 17 eyes in the 22.10~22.70 mm group, 18 eyes in the 22.71~23.12 mm group, and 18 eyes in the 23.13~24.48 mm group. Binary Logistic regression analysis was employed to investigate the factors influencing the occurrence of DR and PDR.RESULTS:Binary Logistic regression analysis showed that both axial length and age significantly influenced the development of DR and PDR.(Axial length: OR=0.296, 95%CI:0.130~0.672, P<0.05; OR=0.237, 95%CI:0.076~0.736, P<0.05; age: OR=0.949, 95%CI:0.907~0.994, P<0.05; OR=0.879, 95%CI: 0.820~0.942, P<0.05). The risk of PDR in the group with axial length of 23.13~24.48 mm was reduced compared to the group with axial length of 20.00~22.09 mm(OR=0.057; 95%CI: 0.006~0.515, P=0.011).CONCLUSION:The findings indicate that longer axial length in patients with type 2 diabetes are associated with a decreased risk of developing DR, as well as a reduced likelihood of DR progressing to PDR. Therefore, a long axial length can be considered a protective factor against DR.

ABSTRACT

Objective To explore the potential targets of triclosan in the treatment of nonalcoholic fatty liver disease(NAFLD) and to provide new clues for the future research on the application of triclosan. Methods The targets of triclosan and NAFLD were obtained via network pharmacology.The protein-protein interaction network was constructed with the common targets shared by triclosan and NAFLD.The affinity of triclosan to targets was verified through molecular docking.Gene ontology(GO) annotation and Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway enrichment were carried out to analyze the key targets and the potential mechanism of action.NAFLD model was established by feeding male C57BL/6J mice with high-fat diet for 12 weeks.The mice were randomly assigned into a model group and a triclosan group [400 mg/(kg·d),gavage once a day for 8 weeks].The hematoxylin-eosin(HE) staining was used for observation of the pathological changes and oil red O staining for observation of fat deposition in mouse liver.Western blotting was employed to detect the protein level of peroxisome proliferator-activated receptor alpha(PPARα) in the liver tissue. Results Triclosan and NAFLD had 34 common targets,19 of which may be the potential targets for the treatment,including albumin(ALB),PPARα,mitogen-activated protein kinase 8(MAPK8),and fatty acid synthase.Molecular docking predicted that ALB,PPARα,and MAPK8 had good binding ability to triclosan.KEGG pathway enrichment showcased that the targets were mainly enriched in peroxisome proliferator-activated receptor signaling pathway,in which ALB and MAPK8 were not involved.Triclosan alleviated the balloon-like change and lipid droplet vacuole,decreased the lipid droplet area,and up-regulated the expression level of PPARα in mouse liver tissue. Conclusion PPARα is a key target of triclosan in the treatment of NAFLD,which may be involved in fatty acid oxidation through the peroxisome proliferator activated receptor signaling pathway.

ABSTRACT

Inflammatory myofibroblastoma (IMT) is a rare solid tumor, and its etiology and pathogenesis are unclear. Crohn's disease is a non-specific intestinal inflammatory disease. The clinical manifestations, laboratory examinations, and imaging examinations of IMT are not specific, making diagnosis difficult. A case of Crohn's disease combined with IMT of abdominal wall was admitted to the Department of Gastroenterology at the Third Xiangya Hospital, Central South University, on Nov. 21, 2017. This patient was admitted to our hospital because of repeated right lower abdominal pain for 4 years. A 6 cm×5 cm mass was palpated in the right lower abdomen. After completing the transanal double-balloon enteroscopy and computed tomographic enterography for the small intestinal, the cause was still unidentified. The patient underwent surgery due to an abdominal wall mass with intestinal fistula on Sept. 12, 2018 and recovered well currently. According to histopathology and immunohistochemistry, he was diagnosed with Crohn's disease combined with IMT. Up to July 2020, the patients still took azathioprine regularly, without abdominal pain, abdominal distension, and other discomfort, and the quality of his life was good.

ABSTRACT

Objective:To study and establish a method for the efficient screening of 14-3-3τ protein inhibitors from natural products, and analyze the sites of their interactions with the 14-3-3τ protein. Methods: The binding activity of natural compounds with the 14-3-3τ protein was tested by the liquid chromatography-fluorescence spectroscopy, and the binding activity of the potential compounds was further verified by the surface plasmon resonance(SPR)technique. The binding sites of the active compounds were predicted by the molecular docking technique. Furthermore, the key binding sites were selected and then validated using amino acid site-directed mutants. Results: A total of 17 different type compounds with potential 14-3-3τ binding activity were screened out from 82 natural products. The binding activi- ties of 10 compounds were verified by the SPR experiments. Then the binding sites of interactions between the 14-3-3τ protein and the 10 compounds were predicted by the molecular docking technology to be mainly at the Arg56, Arg127 and Y128A, dem- onstrate that the binding of five of the 10 compounds with the target protein was associated with the three sites Arg56, Arg127 and Tyr128. Conclusion: The established method is accurate and efficient, which could be used for rapid screening of small molecule 14-3-3τ inhibitors from natural products. The present study provides a reference and a new approach for the rapid screening of 14-3-3τ inhibitors for the new breast cancer therapeutic drugs.

ABSTRACT

BACKGROUND:Vitamin D is a key substance that promotes calcium absorption and maintains the health of skeletal system,but its relationship with bone mineral density remains controversial.OBJECTIVE:To determine the relationship between serum level of vitamin D and bone mineral density in the middle-aged and elderly patients in southern China.METHODS:The patients aged over 50 years old admitted in the clinic and in-patient department of a large-scale 3A hospital from July 2014 to December 2016 were enrolled and the clinical data were collected.Then,the association between the serum level of 25-hydroxy vitamin D (25(OH)D) and bone mineral density (at the lumbar spine,femoral neck and total hip) was analyzed.RESULTS AND CONCLUSION:A total of 1 154 patients (77.2% female) were enrolled with a mean age of (64.62±10.48)years,and the mean serum level of 25(OH)D was (62.07±37.40) nmol/L.The serum level of 25(OH)D decreased with age increasing,and the proportion of vitamin D insufficiency/deficiency in patients aged 50-59,60-69 and ≥ 70 years old was 71.3%,77.0% and 80.1%,respectively.Correlation analysis showed that the serum level of 25(OH)D was not associated with bone mineral density at the lumbar spine,but had a weakly positive correlation with bone mineral density at the femoral neck and total hip (r=0.09 and 0.08,P < 0.01).For patients with vitamin deficiency (25(OH)D ≤ 50 nmol/L),there was a significantly positive correlation between serum level of vitamin D and bone minerat density at the lumbar spine,femoral neck and total hip (r =0.117,0.120 and 0.146,P < 0.01).After subgroup analysis,vitamin D level was not associated with bone mineral density in male,but was significantly associated with that in female (r=0.105,0.135 and 0.171 for the lumbar spine,femoral neck and total hip,respectively,P < 0.01).Therefore,the prevalence of vitamin D deficiency is common in middle-aged and elderly patients in southern China.The serum level of vitamin D is not related to bone mineral density in male,but was positively correlated with that in female.

ABSTRACT

Objective Based on the theory of Service Quality to develop a scale for measuring personal digital assistant satisfaction by nurses and to detect the reliability and validity of this scale. Methods Through the literature research, cross-sectional study and present satisfaction evaluation tool for using personal digital assistant by nurse to build the item pool. The items were selected by 15 experts consultation and the pilot survey of 666 nurses. Results The satisfaction evaluation tool for using personal digital assistant by nurse scale consisted of 41 items;the exploratory factor analysis identified 8 principal factors and explained for 65.22%. Pearson correlation coefficient between each dimension was 0.213-0.684(P<0.01). Pearson correlation coefficient between each dimension and total scale was 0.574-0.798(P<0.01). The Cronbach α coefficient of the scale was 0.928 and test-retest reliability was 0.934. Conclusions The satisfaction evaluation tool for using personal digital assistant by nurse scale has good validity and reliability. It can be used as a tool to measure the satisfaction for using personal digital assistant by nurse.

ABSTRACT

Objective Through the analysis of the status quo of the research on neonatal pressure sore in the relevant nursing research, we can understand the strategy and the problems of the prevention of neonatal pressure sore, which provides reference for future research. Methods This article used bibliometric analysis method to construct literature reading and reviewing database,screening articles in the Chinese biological medical disc (CBMdisc), China Journal Full-text Database (CJFD), China Science Periodical Database(CSPD)and China Science and Technology Journal Database(CSTJ), according to the criteria of inclusion and exclusion. Results A total of 74 articles were integrated into the literature, the first published in China in 2006, after the overall trend of growth, especially since 2011 began to significantly increase. Of which 29 (39.2%) of the literature published in 13 kinds of professional nursing journals. The total number of citations was 1.84, the cohort rate was 39.19%, the total number of citations was 54, and the number of citations per article was 0.73. The total number of citations was 447, and the average number of citations was 6.04. Literature research types are subdivided into 10 categories, of which experienced literature was the most (23.0%). Literature research concentrated on the use of chemical or physical methods to prevent neonatal pressure sores. Conclusions Neonatal pressure sore is paid more and more attention by the nurses, and the policy support for it is acceptable, but the related research needs to be further strengthened in the breadth, depth and comprehensive aspects. It is necessary for us to use scientific research methods, combining with the culture of pediatric and the characteristics of pediatric management in our country, to construct the best practice manual for preventing neonatal pressure sore in China.

ABSTRACT

Objective To explore major risk factors for postoperative biliary tract infection associated with endoscopic retrograde cholangiography (ERC), and to evaluate endoscopic nasobiliary drainage (ENBD) for intervening the infection.Methods A total of 512 patients who underwent ERC at the First People's Hospital of Yunnan Province from January 2010 to June 2016 were enrolled and divided into group A and B randomly.Group A underwent ENBD after ERC while group B without.The incidence rates of biliary tract infection in different causes and lesions were compared between the two groups.Results Among the 512 patients, there were 276 cases in group A and 236 cases in group B.The overall postoperative biliary infection rate was 4.30%(22/512).Patients in group A showed a smaller chance of developing postoperative biliary tract infection than that in group B [1.09%(3/276) VS 8.05%(19/236), χ2=15.00, P=0.000].Malignant biliary obstruction was the most common cause (13.46%, 14/104) and the most common site was hepatic portal (13.43%, 9/67).Conclusion ENBD can ensure smooth drainage of bile duct therefore effectively prevent biliary tract infection after ERC, especially for patients with malignant biliary obstruction and hepatic portal lesion.

The application of MNIS have played an important role in confined the nursing profession, disease management and pharmaceutical management, more users discover problem from different professional perspective, thus the MNIS should be improved for all-round care. This article is consist of the domestic and care information systems development, application and effect, as well as its application in the clinical factors of its research progress in clinical application were reviewed to provide a basis for the development of leadership and decision-making about MNIS, and thus, enhance the mobile application of the effectiveness of care information systems and promoting their improvement.

ABSTRACT

<p><b>Objective</b>To evaluate the effect and safety of phloroglucinol combined with parecoxib on cystospasm after transurethral resection of the prostate (TURP).</p><p><b>METHODS</b>We conducted a prospective randomized case-control study on 98 patients treated by TURP. After operation, the patients were randomly assigned to a treatment (n=50) and a control group (n=48), the former treated by intravenous injection of 80 mg phloroglucinol qd plus 40 mg parecoxib bid while the latter given 80 mg phloroglucinol only, both for 3 successive days. Then we recorded the frequency and duration of cystospasm, visual analogue scales (VAS), adverse reactions, post-operative bladder irrigation time, catheter-indwelling time, and hospital stay and compared them between the two groups of patients.</p><p><b>RESULTS</b>Compared with the controls, the patients in the treatment group showed a significantly lower frequency of cystospasm ([1.95±0.14] vs [0.70±0.65] times, P<0.01), duration of cystospasm ([0.44±0.21] vs [0.12±0.14] min, P<0.01), and VAS score (2.70±1.80 vs 1.90±1.30, P<0.01) at 48-72 hours after TURP, but no statistically significant differences were found between the control and treatment groups in the post-operative bladder irrigation time ([2.75±0.87] vs [2.64±0.83] d, P>0.05), catheter-indwelling time ([3.52±0.32] vs [3.44±0.42] d, P>0.05), and hospital stay ([5.23±0.81] vs [5.10±0.73] d, P>0.05), and no obvious adverse reactions were observed in either of the two groups.</p><p><b>CONCLUSIONS</b>Phloroglucinol combined with parecoxib is more effective and safer than phloroglucinol alone in relieving postoperative cystospasm after TURP.</p>

ABSTRACT

Objective To evaluate the double-balloon enteroscopy(DBE) for obscure gastrointestinal bleeding(OGIB) and evaluate the health economics under the guidance of DBE for surgical treatment of obscure gastrointestinal bleeding.Methods A total of 114 patients,whose hemorrhage etiology could not be determined by conventional gastroscopy,enteroscopy and gastroenterography,underwent DBE.With pathological results as diagnostic criteria,the value of double-balloon enteroscopy for obscure gastrointestinal bleeding diagnosis was studied.Results The sensitivity,specificity and accuracy of DBE for OGIB were 85.86%,63.63%,81.57%,respectively.The positive likelihood ratio was 2.36 and the negative likelihood ratio was 0.22.The total days of hospitalization,hospital costs,cost of hospital drugs were lower in the DBE group than in the control group(27.2 ± 11.8 days VS 16.4 ±5.3 days,35 690.2 ±3 466.5 Yuan VS 19 409.3 ± 9 253.2 Yuan,17 805.8 ± 2 145.5 Yuan VS 9 133.0 ± 4 664.9 Yuan) (all P < 0.05).Conclusion DBE plays an important role in diagnosis of OGIB.It has significance in clinic,health economics,and social benefits.

ABSTRACT

Objective To investigate the expressions of CXC chemokine 10(CXCL10) in chronic non-atrophy gastritis(CNAG) , precancerous lesions(PL)and gastric cancer(GC) ,primitively understanding of CXCL10 expression levels in three gastric types ,ex-ploring their clinical significances .Methods The expressions of CXCL10 in 20 cases of CNAG ,60 cases of PL ,60 cases of GC tis-sues were examined with immunohistochemistry method ,the expression level of CXCL10 was analyzed by computer-assisted image analysis system ,and then analyzed statistically .Results CXCL10 expression were positive in parts of CNAG ,PL and GC the posi-tive rates were 10 .00% ,26 .67% ,71 .67% respectively) .Expression levels of CXCL10 in the GC tissue specimens were significant-ly higher than in CNAG and PL(P0 .05) .Expression levels of CXCL10 in CAG with IM ,CAG with Dys had no significant difference(P>0 .05) ,and in CAG with Severe Dys and Light-Moderate Dys had no significant difference(P>0 .05) .The expression levels of CXCL10 were rele-vant to the differentiation degree of GC (P>0 .05) .Conclusion The expression levels of CXCL10 were gradually rose from CNAG , PL to GC ,and had significant correlation with each other in CNAG ,PL and GC ,indicating that CXCL10 have a key role in the pro-duce and development of GC .

ABSTRACT

<p><b>OBJECTIVE</b>To observe clinical efficacy of plum-blossom needle for follicular maldevelopment (FM).</p><p><b>METHODS</b>Fifty cases of FM were randomly divided into a plum-blossom needle group and a medication group, 25 cases in each one. In the plum-blossom needle group, the plum-blossom needle was applied along Thoroughfare, Conception, Governor and Belt Vessel as well as at Pishu (BL 20), Weishu (BL 21), Shenshu (BL 23), Luanchao (Extra), Zigong (EX-CA 1) during the follicular growth phase, once every other day. In the medication group, clomifene (CC) was prescribed for oral administration and human choriogonadotropin (HCG) was given by intramuscular injection, once each day. For both groups, one menstrual cycle constituted one course. After two courses of treatment, follicular development condition, the changes of endometrial thickness and morphology, ovarian resistent index (RI) and pulsatility index (PI), rate of ovulation and pregnancy were compared between the two groups.</p><p><b>RESULTS</b>After the treatment, the average diameters of the biggest follicle increased in both groups, while the endometrial thickness and morphology in the plum-blossom needle group were superior to those in the medication group (all P < 0.05). Ovarian RI and PI during mature follicular phase in the plum-blossom needle group were inferior to those in the medication group (both P < 0.05). The differences in ovulation and pregnancy rate were not significant statistically between the two groups (both P > 0.05).</p><p><b>CONCLUSION</b>The plum-blossom needle therapy based on regulating Thoroughfare, Conception, Governor and Belt Vessel could improve the ovarian blood perfusion, promote the follicular growth, increase the ovulation rate of mature follicle and avoid the out-of-sync between growth of follicle and endometrium during the treatment of western medication.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the feasibility and safety of patient controlled analgesia (PCA) technology during double balloon endoscopy(DBE) inspection.</p><p><b>METHODS</b>According to the anesthesia, 120 patients with suspected intestinal disease were randomized into non-anaesthesia(Group A), propofol infusion with TCI vein pump(Group B), and remifentanil vein infusion with PCA pump(Group C), with 40 patients in each group. The feasibility and safety of the three methods in double balloon endoscopy(DBE) inspection were evaluated.</p><p><b>RESULTS</b>The tolerance in groups B and C was good and the procedure success rate was 100%, significantly higher than Group A(80%, 12/40, P<0.01). The fluctuation of blood pressure, heart rate, and oxygen saturation was significantly greater in groups A and B, while patients in Group C remained stable. The detection rate was 67.5%(27/40) in Group B and 72.5%(29/40) in Group C, significantly higher than that in Group A(37.5%, 15/40, all P<0.01). The depth of endoscope was longer and the length of hospital stay shorter in Group C as compared to Group B(both P<0.05).</p><p><b>CONCLUSION</b>The application of PCA technology in the double-balloon endoscopy inspection can meet the individualized analgesic needs of patients in different period, increase the tolerance and compliance of patients to the inspection, facilitate patient-operator communication and positions change, reduce the operation risk, and increase the success rate of double balloon endoscopy.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the implication of the dynamic changes of plasma N-terminal pro-B-type natriuretic peptide (NT-proBNP) level and Tei index of left ventricle (LV) in children with ventricular septal defect (VSD) treated by transcatheter closure.</p><p><b>METHODS</b>Sixty children with VSD treated by transcatheter closure with VSD occluder (Group VSD) and 30 healthy children (Group C) were included in this study. The plasma concentration of NT-proBNP, Tei index of LV and left ventricle ejection fraction (LVEF) were measured in Group C and at before, 5th minute, 4th hour, 1st month, 3rd month and 6th month after VSD closure in Group VSD.</p><p><b>RESULTS</b>(1) The concentration of plasma NT-proBNP was significantly increased in children with VSD before transcatheter closure compared with Group C [(229.45 ± 57.75) ng/L vs. (99.21 ± 46.86) ng/L, P < 0.01], significantly increased at 5th minute and 24th hour after transcatheter closure [(356.27 ± 96.78) ng/L and (356.38 ± 91.95) ng/L vs. (229.45 ± 57.75) ng/L, all P < 0.01], and significantly decreased at 1st month, 3rd months and 6th months after transcatheter closure [(131.33 ± 34.79) ng/L, (96.56 ± 31.55) ng/L and (93.39 ± 29.46) ng/L vs. (229.45 ± 57.75) ng/L, P < 0.05 or P < 0.01]. (2) The Tei indexes of LV in Group VSD before transcatheter closure were significantly higher than in Group C (0.45 ± 0.05 vs. 0.33 ± 0.08, P < 0.01) and Tei index was significantly increased at 24th hour, 1st month after transcatheter closure (P < 0.01) while significantly decreased at 3rd and 6th month compared with those before transcatheter closure (0.34 ± 0.07 and 0.34 ± 0.06 vs. 0.45 ± 0.05, all P < 0.01). (3) There is a positive correlation between the changes of the plasma concentration of NT-proBNP and the change of Tei index of LV before and after transcatheter closure (r = 0.653, P < 0.05).</p><p><b>CONCLUSION</b>Tei index of LV and NT-proBNP can monitor cardiac function changes in children with VSD before and after transcatheter closure.</p>

ABSTRACT

TGFβ/smad pathway is recognized as an important signal pathway to promote the pathogenesis of atherosclerosis (AS). Peroxisome proliferator-activated receptor γ (PPARγ) activation is considered to be important in modulating AS. Herein, we investigated the regulation of PPARγ on c-Ski, the repressor of TGFβ/smad pathway, in rat AS model and cultured vascular smooth muscle cells (VSMCs). c-Ski mRNA and protein expression were detected by real-time PCR and Western blot, respectively, in vivo and in vitro with treatment of PPARγ agonist rosiglitazone and antagonist GW9662. The proliferation and collagen secretion of VSMCs after c-Ski transfection were investigated. The underlying mechanism was further investigated by online program NUBIScan and luciferase reporter gene analysis. Results showed that both mRNA and protein expressions of c-Ski in the AS lesions was down-regulated in vivo, while in cultured VSMCs, c-Ski transfection significantly suppressed the proliferation and collagen secretion of rat VSMCs. Rosiglitazone significantly up-regulated mRNA and protein levels of c-Ski in VSMCs, which could be blocked by GW9662. Online NUBIScan analysis suggested possible PPARγ binding sites in the promoter region of c-Ski. In addition, luciferase activity of c-Ski reporter gene was also increased obviously in the presence of rosiglitazone. These results indicate that c-Ski is one of the newly found target genes of PPARγ and thus involved in the anti-AS effect of PPARγ.

ABSTRACT

The aim of this study is to investigate the critical factor affecting the properties of PLGA microspheres fabricated by a solid-in-oil-in-water (S/O/W) emulsion technique with BSA as a model protein. Prior to encapsulation, the BSA microparticles were fabricated by a modified freezing-induced phase separation method. The microparticles were subsequently encapsulated into PLGA microspheres by S/O/W emulsion method, then Motic BA200 biological microscope, confocal laser scanning microscope, scanning electron microscope were used to observe the structure of S/O/W emulsion and PLGA microspheres. The protein content extracted or released from BSA microspheres was measured by Bradford protein assay method. It was found that NaCl added in the outer aqueous phase effectively suppressed material exchange between the inner and outer phase of S/O/W emulsion. Then, the structure and permeability of obtained microspheres were influenced. As a result, with the increase of NaCl concentration in the outer aqueous phase, the encapsulation efficiency of microspheres significantly increased from 60% to more than 85%, the burst release of microspheres reduced from 70% to 20%, and the particle size decreased from 103 microm to 62 microm. Furthermore, the rehydration of encapsulated protein was also retarded and then integrity of BSA was successfully protected during encapsulation process. In vitro release test showed that BSA released from PLGA microspheres in a sustained manner for more than 30 days.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the effects on cardiac remodeling post transcatheter closure by Amplatzer septal occluder selected by oval circumference formula in patients with atrial septal defect (ASD).</p><p><b>METHODS</b>A total of 146 patients with ASD (68 males,mean 33.5 years) treated by transcatheter closure with the Amplatzer occluder were enrolled in this study. The diameter of defects was corrected with the oval circumference formula (group A, 73 cases) or by echocardiography (group B, 73 cases). Cardiac remodeling was assessed by transthoracic echocardiography (TTE) before the procedure, 3 days, 3 months and 6 months after ASD closure.</p><p><b>RESULTS</b>The mean ASD diameter was similar between the two groups [(20.16 +/- 4.98) mm vs. (21.36 +/- 5.69) mm, P > 0.05] and the mean diameter of the selected occluder of group A was significantly smaller than that in group B [(21.95 +/- 6.78) mm vs. (25.85 +/- 6.75) mm, P < 0.05]. Procedural success rate was identical between the two groups (97.3%) and the defects were completely occluded and there was no residual shunt during the 6 months follow up period, there were also no complications during and after the procedure. The lateral diameter of right atrial (RALD), the diastolic diameter of right ventricle (RVDD), RALD/LALD, RVDD/LVDD and pulmonary diameter (PD) were significantly decreased while the lateral diameter of left atrial (LALD) and left ventricle (LVDD) were significantly increased post ASD closure in both groups. At 6 months follow up, RALD decreased by (18.63 +/- 10.59)% in group A versus (10.14 +/- 6.59)% in group B, LALD increased by (13.42 +/- 8.38)% in group A versus (9.28 +/- 4.95)% in group B and RALD/LALD ratio decreased by (26.35 +/- 11.24)% in group A versus (13.98 +/- 8.96)% in groups B (all P < 0.05).</p><p><b>CONCLUSION</b>ASD occluder selection based on the oval circumferen ce formula is superior to that made by echocardiography in terms of more favorable cardiac remodeling post ASD closure.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of FXR on scavenger receptor class B type I (SR-BI) expression.</p><p><b>METHODS</b>Human vascular endothelium Eahy926 cells were treated with FXR agonist androsterone, and the specific target gene of FXR SHP mRNA was detected by RT-PCR. SR-BI mRNA and protein were determined using RT-PCR, real-time PCR and Western blotting.</p><p><b>RESULTS</b>The level of SHP mRNA in Eahy926 cells increased after androsterone treatment at different concentrations for 24 h, demonstrating FXR activation in the cells. RT-PCR, real-time PCR and Western blotting detected increased SR-BI expression at both mRNA and protein levels after FXR activation.</p><p><b>CONCLUSION</b>FXR increases the expression of SR-BI in human vascular endothelium cells.</p>
